How Super P Force 160mg Works

Super P Force 160 mg is a combination drug consisting of 100 mg of sildenafil citrate and 60 mg of dapoxetine. It is generally used in the treatment of erectile dysfunction (ED) with premature ejaculation (PE). The two drugs have two different mechanisms of action. 

Sildenafil citrate acts on the Nitric Oxide (NO)-cGMP pathway. Under normal conditions, on sexual stimulation, the nerve endings in the penile tissues release a chemical called nitric oxide. NO triggers the production of the enzyme guanylate cyclase. This enzyme upregulates the production of cyclic guanosine monophosphate (cGMP) by converting GTP molecules into cGMP. The increased concentration of cGMP leads to the relaxation of smooth muscle tissues in the corpus cavernosum (penile erectile tissue). This results in vasodilation (dilation of blood vessels), which facilitates increased blood flow in the tissue. This process leads to the formation of an erection. 

After some time, there is the release of the enzyme phosphodiesterase-5 (PDE-5), which is responsible for the breakdown of cGMP molecules. As a result, the effects caused by the cGMP are reduced, and the erection weakens further, before reverting to its normal state. 

The sildenafil citrate present in Super P Force 160 mg selectively inhibits the PDE-5 enzyme. This causes improved blood flow in the corpus cavernosum and the effects of cGMPs to persist for a required amount of time. Thus, the individual can sustain an erection for a suitable amount of time.

The dapoxetine in Super Force P 160 mg is used in the treatment of premature ejaculation. Under normal conditions, the ejaculation process is controlled by the serotonin (5-HT) pathways in the brain and the spinal cord. Normally, the ejaculation is controlled by the brain, spinal cord (spinal ejaculation generator (SEG)), and the peripheral nervous system. It is decided based on the competition between excitatory and inhibitory molecules, with serotonin being the inhibitory molecule. 

The nerve cells release serotonin as a part of baseline inhibitory control, which is bound to the receptor on the postsynaptic nerve cells, namely the serotonin receptors (5-HT receptors). This keeps the ejaculation in the “WAIT” mode. There are serotonin transporters (SERT) in the nerve synapses, which transport the serotonin back into their parental nerve cells, thus interfering with the “WAIT” signal.

This process is essential to push the body towards ejaculation, but only after the excitatory signals cross the thresholds of the SEG switch. Under premature ejaculation, the serotonin binding to the 5-HTs is interrupted before this threshold is crossed, leading to premature ejaculation.

Dapoxetine selectively targets this SERT, thus allowing the serotonin molecules to bind to their 5-HT receptors, keeping the body in “WAIT” mode for a sufficient amount of time. As the excitatory signals naturally cross the SEG threshold, the system shifts towards ejaculation.
